Semaglutide protocols typically require "titration"—starting with a very small dose to assess subject tolerance, then gradually increasing.
The Titration Challenge
Standard starting doses in research are often 0.25 mg. See mcg vs mg conversion.
If you have a 5mg vial and add only 1ml of water, your concentration is 5mg/ml.
To get 0.25mg, you would draw 0.05ml (5 Units).
5 Units is a very small amount. It is easy to accidentally draw 6 or 7 units, increasing the dose by 20-40%. Learn reading an insulin syringe to avoid this.
The Dilution Solution
Use more water to make the volume larger and easier to read. Learn how to calculate peptide concentration.
Add 2ml water to 5mg vial.
Concentration: 2.5 mg/ml.
0.25mg Dose = 0.1 ml (10 Units).
10 Units is much easier to measure accurately than 5 Units. Comparison with tirzepatide reconstitution calculator shows similar principles.
